| Definition. A karyotype is the complete, organized picture of an individual's chromosomes — photographed during cell division, then sorted into homologous pairs by size and shape. A normal human karyotype is written 46,XX (female) or 46,XY (male). | Definicja. Cells (from blood, or fetal cells from amniotic fluid) are grown in the lab, stopped at metaphase — when chromosomes are at their fattest and most visible — stained to give banding patterns, photographed, and arranged: autosome pairs 1–22 from largest to smallest, sex chromosomes last. What it reveals: wrong counts (an extra or missing chromosome) and large structural changes (a missing piece, a fragment moved onto another chromosome). | Szczegóły. A karyotype cannot see genes: cystic fibrosis, sickle cell, Huntington's — every single-gene disease looks perfectly normal on a karyotype, because a mutation of a few DNA letters is millions of times too small for a microscope. W polskich podręcznikach kariotyp definiuje się jako kompletny zestaw chromosomów komórki somatycznej, charakterystyczny dla gatunku, a jego graficzne uporządkowanie w pary nazywa się kariogramem (idiogramem). Kariotyp człowieka to 46 chromosomów: 22 pary autosomów i para heterochromosomów (XX/XY); chromosomy klasyfikuje się według wielkości i położenia centromeru (chromosomy metacentryczne, submetacentryczne, akrocentryczne). Badanie kariotypu wykonuje się na chromosomach metafazowych limfocytów lub komórek płodowych (amniopunkcja, biopsja kosmówki) i służy ono wykrywaniu mutacji liczbowych (aneuploidie: zespoły Downa, Edwardsa, Patau, Turnera, Klinefeltera) oraz dużych mutacji strukturalnych (delecje, translokacje).
Na maturze najczęstsze polecenia to: odczytaj z kariogramu płeć i zespół chorobowy, zapisz kariotyp (np. 47,XY,+21; 45,X), wyjaśnij, dlaczego badanie wykonuje się na komórkach w metafazie (maksymalna kondensacja chromatyny), oraz rozróżnij pojęcia kariotyp–genotyp–genom.

AP Biology deep dive¶
Resolution defines the tool. Classic G-banding resolves ~5–10 Mb — whole-arm events, aneuploidies, large deletions/translocations — which is why karyotyping catches trisomy 21 but not a 3-bp CFTR deletion. The modern ladder of cytogenetics descends in resolution steps: FISH paints specific loci (~100 kb), chromosomal microarray detects sub-microscopic copy-number variants (~10–100 kb), and sequencing reads single bases. Choosing the right rung for a clinical question ("recurrent miscarriage" vs "suspected point mutation") is a genuinely AP-style experimental-design judgment.
Balanced vs unbalanced. Karyotypes distinguish changes that alter dosage (unbalanced: trisomies, deletions — phenotypic) from rearrangements that only move material (balanced translocations, inversions — often phenotypically silent carriers). The catch: balanced carriers make unbalanced gametes at high rates, because translocated chromosomes pair awkwardly in meiosis I — the mechanism behind familial Down syndrome (Robertsonian 14;21) and much recurrent pregnancy loss. This is meiosis logic applied to a lab image.
Karyotype evolution. Chromosome counts vary freely across species (humans 46, chimps 48, dogs 78) with no complexity meaning; human chromosome 2's fusion — matching two ape chromosomes band-for-band, with relic telomere sequence at the fusion point and a defunct second centromere — is a flagship evidence-of-descent argument. Sex-chromosome systems (XY, ZW in birds, haplodiploidy in bees) show the same machinery solving sex determination differently.
Where it sits in AP. Karyotype analysis supports chromosomal inheritance and nondisjunction in Topic 5.6 of AP Unit 5 (Heredity); the chromosome-2 fusion features in Unit 7 (Natural Selection) as evidence for common ancestry.
