| Definition. The endoplasmic reticulum (ER) is a network of membrane channels and sacs spreading out from the nuclear envelope — part factory floor, part highway. It comes in two connected regions: rough ER, studded with ribosomes, and smooth ER, without them. | Definicja. | How it works. On the rough ER, attached ribosomes push newly made proteins into the channels; inside, the proteins are folded, checked, and wrapped in little membrane bubbles (vesicles) headed for the Golgi apparatus — this is the route for proteins meant for export, for membranes, or for lysosomes. The smooth ER does different chemistry: it builds lipids, helps detoxify drugs and poisons, and stores calcium (crucial in muscle cells). | Jak działa. Siateczka śródplazmatyczna (retikulum endoplazmatyczne, ER) to system kanałów, cystern i pęcherzyków otoczonych pojedynczą błoną, łączący się z otoczką jądrową. Wyróżnia się siateczkę szorstką (granularną), pokrytą rybosomami, oraz gładką (agranularną), pozbawioną rybosomów. Siateczka szorstka uczestniczy w syntezie i obróbce białek przeznaczonych „na eksport”, do błon i do lizosomów; gładka odpowiada za syntezę lipidów, unieszkodliwianie substancji toksycznych i magazynowanie jonów wapnia.
W podręcznikach ER otwiera temat systemu błon wewnętrznych komórki: białko powstałe na siateczce szorstkiej wędruje w pęcherzykach do aparatu Golgiego, a stamtąd do miejsca przeznaczenia. AP Biology deep dive¶
Cotranslational import. What sends a ribosome to the ER is the protein itself: a signal sequence at the start of the growing chain is recognized and docked to the ER membrane, and the rest of the protein threads into the lumen as it is translated. There, chaperones fold it, and glycosylation — the first sugar-tagging step — begins. Misfolded proteins are held back and degraded; when they pile up, the unfolded protein response expands ER capacity or, failing that, triggers apoptosis. The ER lumen is continuous with the space between the two nuclear envelope membranes — one interconnected compartment.
Smooth ER portfolios. Smooth ER synthesizes phospholipids and steroids (steroid-hormone-producing cells in gonads and adrenal cortex are packed with it), detoxifies hydrophobic drugs via cytochrome P450 oxidases (chronic exposure induces more smooth ER — the cellular basis of drug tolerance), and, as the sarcoplasmic reticulum of muscle, sequesters and releases the Ca²⁺ pulses that trigger contraction.
First station of the endomembrane system. Everything the secretory pathway ships — membrane proteins, secreted enzymes, lysosomal hydrolases — starts in the ER and moves on by vesicle traffic to the Golgi. AP expects the ordered pathway: ribosome → rough ER → vesicle → Golgi → vesicle → plasma membrane or lysosome.
