Definition. A DNA fingerprint (DNA profile) is a pattern of DNA fragments that is practically unique to one person — the "read" step of the biotech toolbox, used to match a sample to an individual. How it works. The profile does not read your whole genome. It looks at a dozen-plus special spots where a short DNA phrase repeats itself — short tandem repeats (STRs) — and how many times it repeats differs from person to person. The lab copies those spots with PCR, separates the copies by size using gel electrophoresis, and reads off the band pattern. Match the pattern at all the tested spots and the odds of a random coincidence fall below one in billions. Jak działa. Profil nie odczytuje całego genomu. Example. A hair at a crime scene yields a profile matching the suspect's — or, just as importantly, not matching, which has cleared hundreds of wrongly convicted people. The same technique settles paternity tests (a child's bands must come half from mom, half from dad) and identified victims after disasters. Przykład. Why it matters. DNA profiling rebuilt forensics, family law, and even history (identifying remains of the Romanovs). Common mistake. A DNA fingerprint is not your genome sequence and says nothing about your traits — the tested repeats are chosen precisely because they are not genes. Identical twins share one profile, and a match is a statistics statement ("1 in a billion"), not magic certainty: contamination and mixed samples are real courtroom issues. Częsty błąd. Metoda opiera się na sekwencjach mikrosatelitarnych (STR) — niekodujących odcinkach DNA o zmiennej osobniczo liczbie powtórzeń; do analizy śladowych ilości materiału (włos, ślina, krew) konieczne jest wcześniejsze powielenie DNA techniką PCR (łańcuchową reakcją polimerazy), a rozdział fragmentów zapewnia elektroforeza. Historycznie technikę opracował Alec Jeffreys (1984 r.), a pierwsze głośne zastosowania dotyczyły spraw kryminalnych i testów ojcostwa.

Na maturze typowe zadania każą odczytać wynik elektroforezy: wskazać, które prążki dziecka pochodzą od matki, a które od domniemanego ojca, albo który z profili odpowiada śladowi z miejsca zdarzenia. Warto pamiętać sformułowanie, że profil ustala się na podstawie DNA pozagenowego — dzięki temu nie ujawnia on informacji o cechach czy chorobach badanej osoby.

Why STRs vary so much. Tandem repeats mutate by replication slippage: DNA polymerase stutters on repetitive templates, adding or dropping repeat units at rates far above point-mutation rates. That generates many alleles per locus (high heterozygosity), and because each person carries two alleles per STR, a 13–20-locus panel multiplies exclusion probabilities into the 10⁻¹⁵ range. The loci are deliberately intergenic or intronic — maximally informative for identity, minimally informative about phenotype, a designed-in privacy feature.

The pipeline is this unit's toolbox in one assay. PCR amplifies each STR with fluorescent primers (tiny samples suffice — a licked stamp works); capillary electrophoresis sizes the products to single-base resolution; a child's profile shows one maternal and one paternal allele at every locus — Mendelian segregation, visualized. Older RFLP fingerprinting (restriction digest + Southern blot) is the historical bridge from restriction enzymes to modern profiling.

Statistics and its edge cases. Match probability comes from allele frequencies via the product rule across independent loci — Hardy–Weinberg in forensic clothing, with corrections for population substructure. Live controversies worth knowing: familial searching (partial matches implicating relatives), forensic genetic genealogy (the 2018 Golden State Killer case used a consumer-genealogy database), and degraded or mixed samples where probabilistic genotyping replaces a clean yes/no.
