| Definition. Transfer RNA (tRNA) is the cell's delivery molecule: it brings the correct amino acid to the ribosome during translation. | How it works. The tRNA folds into a compact cloverleaf-like shape. When its anticodon pairs with the matching codon on the mRNA inside the ribosome, the amino acid it carries is added to the growing protein chain. W polskich podręcznikach tRNA opisuje się przez jego charakterystyczną strukturę „liścia koniczyny”: ramiona z pętlami, w tym pętla antykodonowa z antykodonem, oraz koniec 3′ z sekwencją CCA, do której wiązany jest aminokwas. Połączenie tRNA z aminokwasem — aminoacylo-tRNA — powstaje przy udziale enzymu syntetazy aminoacylo-tRNA i wymaga energii z ATP. W translacji tRNA dostarcza aminokwasy do rybosomu w kolejności wyznaczonej przez kodony mRNA.
Na maturze tRNA pojawia się w pytaniach o role trzech rodzajów RNA, w zadaniach sekwencyjnych (dopisz antykodony do podanych kodonów) oraz w poleceniach typu „wyjaśnij, dlaczego tRNA nazywany jest adaptorem/tłumaczem”. AP Biology deep dive¶
An adaptor, as predicted. Crick reasoned in the 1950s that nucleic acids cannot recognize amino acids directly, so an "adaptor" must exist — tRNA proved him right. The cloverleaf secondary structure folds further into an L-shape: anticodon loop at one tip, the amino acid esterified to the 3′-CCA end at the other, ~70–90 nucleotides with unusual modified bases throughout. The two functional ends are far apart, which is exactly what the ribosome's geometry requires.
Charging is where fidelity lives. Each of the twenty aminoacyl-tRNA synthetases recognizes its amino acid and all tRNAs for it, couples them using ATP (the aminoacyl bond stores the energy later spent on the peptide bond), and several proofread — an editing site hydrolyzes near-miss amino acids like valine mistakenly loaded for isoleucine. The genetic code's accuracy is therefore enforced twice: once at charging, once at codon–anticodon pairing in the ribosome.
Wobble economics. Cells run ~45 tRNA species against 61 sense codons because the anticodon's first position (reading the codon's third) pairs loosely — inosine there can read U, C or A. This matches the code's degeneracy pattern and buffers many third-position mutations into silence. During elongation, each tRNA transits the ribosome's A → P → E sites: arriving charged, holding the growing chain, leaving empty — a courier's loop run millions of times per cell per minute.
