W polskich podręcznikach antykodon opisuje się przy budowie tRNA: to trójka nukleotydów w pętli antykodonowej, po przeciwnej stronie cząsteczki niż miejsce przyłączenia aminokwasu (koniec 3′). Antykodon jest komplementarny i antyrównoległy względem kodonu mRNA — a ponieważ każdemu antykodonowi odpowiada ściśle określony aminokwas, tRNA działa jak „tłumacz” między językiem nukleotydów a językiem aminokwasów.
Typowe zadanie maturalne to uzupełnianie łańcucha zapisu: nić matrycowa DNA → kodony mRNA → antykodony tRNA → aminokwasy. Uwaga na pułapkę: antykodon ma tę samą sekwencję co odpowiadający fragment nici matrycowej DNA, tylko z uracylem zamiast tyminy — łatwo więc „przeskoczyć” o krok i wpisać sekwencję kodonu zamiast antykodonu. AP Biology deep dive¶
Wobble: fewer tRNAs than codons. Cells do not keep 61 tRNA types for 61 sense codons — humans manage with roughly 45. The wobble hypothesis explains how: the third codon position pairs loosely, so one anticodon (often using the modified base inosine) can read several codons differing only in that position. Wobble maps neatly onto the code's degeneracy — the "sloppy" pairing position is exactly where synonymous codons differ, so flexibility there costs no accuracy in the protein.
The real enforcers of the code are enzymes. The anticodon does the pairing, but nothing about a tRNA's shape forces the right amino acid onto it. That fidelity comes from the twenty aminoacyl-tRNA synthetases — one per amino acid — each of which recognizes both its amino acid and its tRNA set and joins them using ATP, some with proofreading pockets that reject near-miss amino acids. In a classic experiment, an amino acid chemically altered after charging was inserted wherever the tRNA's anticodon dictated: the ribosome trusts the anticodon and never re-checks the cargo.
Geometry checked by the ribosome. In the A site, the small subunit's rRNA probes the codon–anticodon helix and lets GTP-driven proofreading reject poor matches, holding translation errors to about 1 in 10⁴. Accuracy is thus layered: synthetase charging, wobble-limited pairing, and ribosomal inspection — three checkpoints between a codon and its amino acid.
